i.Mune Treg [RUO] ist ein quantitativer Test zur Bestimmung der relativen und absoluten Anzahl menschlicher FOXP3/TSDR+ regulatorischer T-Zellen (Treg) und CD3+ T-Zellen in flüssigem Vollblut oder Trockenblut.

(die absolute Quantifizierung steht zur Zeit nur für flüssiges Vollblut - frisch oder gefroren - zur Verfügung)

Ermöglicht die epigenetische Quantifizierung von1):

  • FOXP3/TSDR+ regulatorischen T Zellen (Treg)
  • CD3+ T-Zellen

TSDR = Treg Specific De-methylated Region


Die bevorzugte Methode zur Quantifizierung von FOXP3+ Treg4)

FOXP3 TSDR is de-methylated in Treg and fully methylated in stimulated CD4+ T cells, that transienly express the FOXP3 protein (Baron et al., 2007)

Der Anteil der Tregs an den Gesamt CD4+ Zellen ist vergleichbar zwischen der FOXP3-Durchflusszytometrie (A, 10,4%) und der TSDR-Demethylierungsanalyse (B, 12,7%).
Stimulierte CD4+ T-Zellen exprimieren transient das FOXP3-Protein (A, 29,7 %), weisen aber keine TSDR-Demethylierung auf (B, 1,4 %). Daher ist die TSDR-Demethylierungsanalyse für die Treg-Quantifizierung spezifischer als der FOXP3-Nachweis mittels Durchflusszytometrie1)4)

  • IPEX Syndrom (immune dysregulation, polyendocrinopathy, enteropathy, X-linked inheritance)3)
  • Andere primäre immunregulatorische Erkrankungen4)

  • Allergien und Asthma6)
  • "Graft-vs-host disease" nach Stammzelltransplantation7)
  • Organ Transplantationen, Tumor Progression, Immuntherapie, Schwangerschaft8)-10)

**Die oben genannten klinischen Anwendungen wurden mit Technologien durchgeführt, die derzeit in der klinischen Laborroutine eingesetzt werden (z. B. Durchflusszytometrie). Epimune hat die Gleichwertigkeit seiner Methode zur Quantifizierung epigenetischer Immunzellen mit der Durchflusszytometrie (in Vollblut) und die Konkordanz der Quantifizierung epigenetischer Immunzellen zwischen Vollblut und Trockenblut nachgewiesen1)
